How is it possible to isolate exosomes by ultracentrifuge?

Since there must be substances which has the similar mass with exosomes,  won't using ultracentrifugation get exosomes and also other micro stuffs?

There are ways to reduce the amount of contaminating materials though, such as 200nm filtration and density gradient ultracentrifugation.

that is true. the ultracentrifugation procedure overall is still one top ones in terms of final exo purity. One should be careful with filtration though- some material is lost along the way. and exosomes are damaged upon prolonged centrifugation at >100,000g
yes, despite ultracentrifugation is considered a gold standard- its not a perfect procedure. with straight sedimentation you'll precipitate many other vesicles and proteins along with exosomes. and with sucrose gradient you'll still recover vesicles that have the same density (M/V) as exosomes. some of them will be way bigger than expected (exosomes should be ~30-150 nm)
